(A) RAW264.7 cells (5 × 103) were seeded, then RANKL (50 ng/mL) with or without the indicated concentration of BAG (5–40 mM) was placed in the osteo-surface well for 5 days. The percentage of bone resorption pit area relative to the total osteo-surface well area was quantified for the experimental condition. RAW 264.7 cells were cultured in the RANKL with or without BAG for 1, 3, and 5 days to induce osteoclast differentiation, and the protein expression of integrin β3 was measured by Western blot (B), and the levels of CCL4 mRNA were measured by real-time PCR (C).
