The NRF2-sMaf heterodimer regulates cytoprotective gene expression through the CNC-sMaf-binding element (CsMBE) motif. (A) NRF2 forms a heterodimer with sMaf and binds to the CsMBE motif in the nucleus, which is classically referred to as an antioxidant responsive element (ARE)/electrophile responsive element (EpRE) motif. NRF2 positively regulates genes encoding detoxicating enzymes and antioxidative enzymes and negatively regulates genes coding for proinflammatory factors. Ub, ubiquitin. P, phosphorylation. (B) The combination of an E3 ligase and the adaptor for the degradation of NRF2 through ubiquitination: KEAP1–Cul3, βTrCP–Cul1 and WDR23-Cul4 complexes.
