Detection of lysine isobutyrylation on protein lysines in 293T cells. Cellular proteomes and core histone proteins were extracted and tested with anti-butyryllysine antibody (PTM Biolabs, Cat#PTM-301). Treatment of cells with isobutyrate induced increase of lysine isobutyrylation level on core histone proteins while no appreciable change was observed on non-histone protein isobutyrylation upon isobutyrate treatment.
