Proposed model for global and gene-specific regulation by Pus6 and MetRS. Pus6 modifies uridines to pseudouridines on both elongator tRNAMet and target mRNAs (e.g. YEF3). These modifications enhance MetRS binding on both types of RNA. MetRS binding to tRNAMet presumably affects its charging and thereby has a global impact on translation, while MetRS binding to an mRNA leads to a gene-specific impact.
