Cyclic tensile strain inhibits IL-1β mediated NO and PGE2 release via a TRPV4 dependent pathway in isolated chondrocytes. The TRPV4 antagonist, GSK205 (10 μM), abolishes the anti-inflammatory effects of mechanical loading (CTS, 10%, 0.33 Hz) at 1 ng/ml IL-1β on (A) nitrite and (B) PGE₂ release at 24 h. Data represents mean ± SD for n = 6 wells per group using cells isolated from 2 different donors.
