High doses of UT prevented entry into phase M promoting cdc2 phosphorylation and inactivating the Cyclin B/cdc2 complex. (a) C2C12 cells were treated with a mixture of 100 µg/mL IS and 100 µg/mL PC (UT) for 24 h in the presence or absence of 0.1 mg/mL colcemid (Co). Tubulin (red) and Histone H3 (green) expression and DAPI (blue) were analysed by immunofluorescence by using a confocal microscope. A representative experiment with negative staining control is shown. (b) C2C12 cells were incubated with a mixture of 100 µg/mL IS and 100 µg/mL PC (UT) or vehicle (CT) for 24 or 48 h in the presence or absence of 0.25 mM probenecid (PB). Cyclin B and phospho-cdc2 and cdc2 expression were analysed by western blotting. Representative blots are shown. Full-length blots are presented in Supplementary Figure S3.
