Both proteins contain an N-terminal signal peptide sequence (SS), a conserved extracellular TMEM132D_N and a TMEM132 domain, a transmembrane (TM) domain. The Drosophila dtn protein contains an intracellular coiled-coil (CC) domain that is missing in human TMEM132 proteins. The structure prediction was made using the SMART (http://smart.embl-heidelberg.de/) protein sequence analysis tool. Arrows point to the positions on the peptide where mutations were generated via CRISPR/Cas9 in this work.
