miR-34a suppresses SIRT1 mRNA expression by directly binding to its 3'-UTR. miR-34a mimics and inhibitors were transfected into human umbilical vein endothelial cells to change following which the effects of transfection on the level of (A) miR-34a and (B) SIRT1 mRNA expression were verified using reverse transcription-quantitative PCR. (C) Changes in the level of SIRT1 protein expression following transfection with the miR-34a mimic or miR-34a inhibitor were measured using western blot analysis and (D) quantified.
