Type I and type II collagen immunofluorescence in pellets treated with PTHrP. After 3 weeks of culture in chondrogenic media pellets were fixed in 10% v/v formalin, sectioned, and probed with antibodies to type I collagen (COL I) and type II collagen (COL II). Sections were imaged by fluorescent microscopy. The cell source nasal chondrocytes (NC), bone marrow-derived mesenchymal stem cells (BM-MSC), or coculture (CC) and PTHrP treatment is indicated at the top of the images. COL I is shown in red, COL II is shown in green, and DAPI is shown in blue in the merged images. (A–F) Immunofluorescence images of pellets associated with interaction indices > 1 (i.e., Responders). (G–L) Immunofluorescence images from pellets associated with interaction indices < 1 (i.e., Non-Responders). All pellets here were formed from 250,000 cells.
