Speculative model of AP-5/SPG11/SPG15 recruitment. We propose that the AP-5/SPG11/SPG15 complex binds to two molecules on the late endosomal or lysosomal membrane: PI3P, via the SPG15 FYVE domain, and RagC in its GDP-loaded state, via SPG11 and/or SPG15. The strong inhibitory effect of RagAGTP and RagBGTP suggests that RagA/B also needs to be in its GDP-loaded state.
