HT-1376 and T24 cells were treated with GlcNAc (4 mM), PuGNAc (25 mM) or GlcNAc + PuGNAc for 24 h, then Chx ((100 µg/mL) was added to stop protein synthesis. (A) Then cells were lysed and equal amounts of protein were separated to WB to measure NUSAP1 expression. (B) HT-1376 and T24 cells were transfected sh-OGT and sh-NC for 24 h, then cells were collected and analysis for RT-PCR to determine the mRNA level of OGT (n=3, ***P<0.001, sh-OGT group vs sh-NC group).
