Fig. 2 — LUBAC exists in distinct complexes with the DUBs OTULIN (1) and CYLD-SPATA2 (2). The LUBAC–DUB interactions are mediated by the PUB domain in HOIP and PUB-interacting motifs in OTULIN and SPATA2. The two LUBAC–DUB complexes can both generate Met1-Ub but have distinct DUB activities; OTULIN disassembles exclusively Met1-Ub whereas CYLD disassembles Met1-Ub and Lys63-Ub.
