a In vitro luciferase assay was performed to determine the effect of mutations of Sp1 binding sites on the activity of TERT promoter in the indicated cells. wtSp1 wild-type Sp1 binding sites, mSp1 mutant Sp1 binding sites, mTERT TERT promoter with C250T mutation, wtTERT wild-type TERT promoter. Mutant (b) and wild-type (c) TERT promoter-driven luciferase constructs with mutant or wild-type Sp1 binding sites were transfected into B16F10 and MC38 cells, and the ChIP-qPCR assays were then performed to determine the effect of the presence of Sp1 binding sites on the recruitment of Sp1/GABP to TERT promoter in the indicated cells.
