lncRNA CRNDE recruits enhancer of zeste homolog 2 (EZH2) and suppresses the expression of p21. (A) RT-qPCR was performed to measure the level of CRNDE in nuclear and cytoplasmic fractions of A549R and H1299R. (B) RIP assay was performed to confirm the combination of CRNDE and EZH2. (C) The cell cycle-related target of EZH2 was measured in LAD radioresistant cells in response to CRNDE knockdown. (D) The level of p21 both in mRNA and protein was detected in response to EZH2 silencing. (E) Chromatin Immunoprecipitation (ChIP) assay revealed that CRNDE could recruit EZH2 to the promoter region of p21 and induces histone H3 lysine 27 trimethylation (H3K27me3) modification. All data are represented as the mean ± SD from three independent experiments. The p value represents the comparison between groups (**p < 0.01).
