Effects of RDV and its parent nucleoside GS-441524 on mitochondrial protein synthesis after 5 days of treatment in PC-3 cells. The effects on COX-1 levels (● blue, solid lines), SDH-A (♦ green, solid lines), and ATP levels (■ red, solid lines) are shown as % of the DMSO control. RDV showed nonselective inhibition of COX-1, SDH-A, and ATP levels, while GS-441524 showed no effect up to 100 μM. The positive control chloramphenicol, a known mitochondrial toxin, specifically inhibited COX-1 synthesis.
