Effects of (+)-betulin on zebrafish. (A) Toxicity on zebrafish development. Embryos at 24 hpf were treated for 24 h and 48 h with (+)-betulin at 50 μM or cycloheximide as the positive control or water as control. (B) Effects of (+)-betulin on the NF-κB pathway in zebrafish. Animals at 72 hpf were treated with betulin at 0, 0.8, 4.5 and 22.5 μM or rocaglamide (3μM) for 5 h. Western blotting was performed to evaluate IKKα and β, NF-κB p50 and p65 expression, and caspase-3 and −7 expression. β-actin levels were used as internal controls.
