Fgd5 GEF assays (a) and GST-Rac1 binding assays (b) were performed in the presence of varying concentrations of aurintricarboxylic acid. (a) Aurintricarboxylic acid inhibited Fgd5 Rac1 GEF activity with a calculated IC50 of 157 nM. Error bars = s.e.m., n = 4. (b) The IC50 for inhibition of GST-Rac1 binding was 1.8 μM for the Fgd5 DH-PH domain (upper panel), and 1.1 μM for the Fgd5 DH-PH-FYVE domain (middle panel), while no inhibition was observed for binding the TrioN DH-PH domain (lower panel)
