URA Inhibited the Activation of Dendritic Cells and Microglia in CNS. Mice were treated with vehicle or URA (25 mg/kg) on the day of EAE induction and sacrificed at day 20 p.i. Spinal cords and brains were harvested and mononuclear cells (MNCs) isolated, following stimulation with MOG35–55 (25 μg/mL) for 24 h. (a) Percentage of CD11c+ cells infiltrated into CNS in URA- or vehicle-treated group. Frequencies of CD11c+ CD80+ (b), CD11c+ CD86+ (c), CD11c+ CD40+ (d), CD11c+ CD14+ (e) cells were assessed by flow cytometry. (f) Absolute numbers of infiltrated CD11c+ cells were calculated by multiplying the percentages of these cells with total numbers of MNCs in each group. (g) The percentage of CD45high CD11b+, CD45low CD11b+, and CD11b+ MHCII+ (h), CD11b+ TNF-α+ (i), CD11b+ CD16/32+ (j) cells in Vehicle- or URA-treated EAE mice.
