Koumine (KM) attenuates MSU-induced NLRP3 inflammasome activation in vivo. C57BL/6 mice were intraperitoneally pretreated with KM (0.8, 2.4, and 7.2 mg/kg), MCC950 (10 mg/kg) or vehicle for 1 h and then given an intraperitoneal injection of MSU crystals (1 mg dissolved in 500 µl of PBS). Six hours later, (A,B) the frequency of CD11b+Ly6G+ neutrophils retrieved from the peritoneal lavage fluid was analyzed by flow cytometry. (C) The amounts of IL-1β in the lavage fluid were quantified by ELISA. The data are representative of those obtained from eight mice in each treatment group.
