IFN-γ production by CD4 T cells is not altered by treatment with tofacitinib. Mice were infected with Leishmania and 2.5 weeks later daily i.p. treatment with tofacitinib or vehicle started. (a) Ear thickness was measured weekly and at 5 weeks (b) parasite numbers (c) and IFN-γ production by CD4 T cells was assessed by flow cytometry. Cells from (d) infected ears or (e) draining LN (dLN) were stimulated with PMA and ionomycin in the presence of brefeldin A for 4 hours and analyzed by flow cytometry for the expression of intracellular IFN-γ.
