Cytokine release in the presence of plasma and BMDM from WT and CETPTg mice . Bone-marrow-derived macrophages (BMDM) were isolated and pooled from WT (gray, n = 9) or CETPTg mice (black, n = 9) and cultured as described under Materials and Methods. Differentiated cells were treated in the presence of increasing amounts of LPS (10, 100, or 1,000 ng/mL) in the presence of 10% serum from either WT (n = 9, hatched bars) or CETPTg mice (n = 8, filled bars) for 2, 6, or 24 h. BMDM response to LPS was assessed by measuring levels of secreted IL-6 (A), MCP-1 (B), IL-10 (C), TNFα (D), and IL-1β in the supernatants by multiplex assay.
