Real-time PCR and ELISA measurements of hepcidin expression in lipopolysaccharide (LPS)- and lipoteichoic acid (LTA)-treated THP-1 cells. Real-time PCR was performed with SYBR Green protocol. β-actin was used as internal control for the normalization. The relative expression of untreated controls was regarded as 1. The mRNA expressions of the treated cells were compared to their appropriate controls (6 h and 24 h). Secreted hepcidin concentrations of the culture media were determined with a human hepcidin ELISA kit according to the instructions of the manufacturer.
