Figure 2

The protective function of exosomes on chondrocytes. (A) Uptake of exosomes by chondrocytes. (B) Effects of IL-1β and exosome treatment on chondrocyte viability were confirmed by MTT assay. (C) Effects of IL-1β and exosome treatment on DNA synthesis activity of chondrocytes were confirmed by EdU assay. (D) Effects of IL-1β and exosome treatment on apoptosis rate of chondrocytes were confirmed by flow cytometry. (E) The protein expression of Collagen Ⅱ and MMP13 in chondrocytes was detected by Western blot assay. All the experiments were performed independently in triplicate and the results were the mean of three values. One-way ANOVA (AD) or two-way ANOVA (E) was used for comparison among multiple groups. *p < 0.05 vs 6 h; #p < 0.05 vs control group; @p < 0.05 vs OA group.

Abbreviations: MTT, 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium; EdU, 5-ethynyl-2′-deoxyuridine; MMP13, matrix metalloprotease 13; ANOVA, analysis of variance; OA, osteoarthritis.
