(a) Estimated occupancy of samples analyzed with Orbitrap Fusion Lumos mass spectrometer based on available quantification values of (glyco)peptides. Circle diagrams depict proportions of O-glycosylated (yellow) and non-O-glycosylated (grey) peptides based on integrated MS1 signals of relevant peptides. When in close vicinity to predicted N-glycosites, separate quantifications for non-N-glycosylated peptides (N(Asn)-peptides) and presumably N-deglycosylated deamidated peptides (D(Asp)-peptides) after PNGase F treatment are provided. Coverage by peptides is shown under each graph. (b) Peptides covering individual O-glycosites indentified with Q Exactive HF-X mass spectrometer were grouped based on length and occupancies based on integrated MS1 signals calculated for each group. Average occupancies of individual O-glycosites -SEM are shown. (c) The cartoon suggests a model, where the majority of identified O-glycopeptides are not N-glycosylated.
