A force pipette (fp) and a pull pipette (pp) are used to extend the nucleus (nuc) (B) A single isolated nucleus can be measured before and after biochemical treatment, providing fine-tuned control over nuclear chromatin mechanical and imaging measurements. The nuclear spring constant (knuc) increases during chromatin condensation and decreases after chromatin digestion.
