LH induces dynamic actin remodeling in BC cells. (A) BC cells were treated for different times (0–60 min) with LH (5 mIU/ml). The total cell amounts of wild-type cortactin and phosphorylated cortactin (cortactinY466) are shown by western blot analysis. (B) Phospho-cortactinY466 densitometry values were adjusted to cortactin intensity and then normalized to the control sample. *P < 0.05 vs. control. (C) T-47D cells were treated with LH for 20 min in the presence or absence of a siRNA paxillin or cortactin3YF construct. Cells were stained with phospho-cortactinY466 linked to FITC, actin was stained with phalloidin linked to Texas Red, and nuclei were counterstained with DAPI. Yellow arrows indicate membrane-localized p-cortactinY466. (D) Quantification of the membrane-localized p-cortactin in the different conditions. Results are expressed as dots/cells (mean ± SD). *P < 0.05 vs. CON, control. Membrane-localized p-cortactinY466 was counted in 40 different cells. All experiments were performed in triplicate, and representative images are shown.
