Kinetics of interaction between EDANS-DnaK and TMR-IAPP. (A) Kinetics of intermolecular FRET between EDANS-DnaK (150–600 nM) and TMR-IAPP (2 μM) monitored using donor (EDANS) fluorescence is shown. (B) Kinetics of aggregation of TMR-IAPP monitored by fluorescence of TMR in presence and absence of 300 nM EDANS-DnaK is shown. Comparison of the kinetics between (A and B) indicates that the interaction occurs in the early phase of aggregation. The solutions are prepared in PBS buffer containing 1 mM EDTA and 1 mM sodium azide at pH 7.4 and incubated at 37°C with continuous stirring. These experiments have been performed twice.
