DNA synthesis reactions containing 10 nM DNA running start substrate with increasing concentrations of Polλ were carried out as described in the Materials and Methods section. A schematic representation of the DNA substrate is shown on top, where TT indicates the position of cyclobutane pyrimidine dimer (CPD) in the damaged template. Reactions in lanes 1–6 contained the undamaged DNA substrate, and reactions in lanes 7–12 contain the DNA substrate harboring a cis-syn TT dimer (CPD). Lanes 1 and 7 contained no enzyme, whereas lanes 2–6 and lanes 8–12 contained increasing concentrations of Polλ (0.2, 0.5, 2.5, 10, and 40 nM, respectively). The position of the primer and of the unmodified TT sequence or the corresponding TT’s of the CPD in the reaction products is indicated on the right.
