Fig. 3. LINC01569 is a candidate mediator of glucocorticoid-induced counteraction of cellular mechanical responses.

Fig. 3

(A) Microarray analysis (GSE151153) of differentially expressed lncRNAs in TA-treated and untreated stretched HSFBs. Red shading indicates increased gene expression, and blue shading indicates decreased expression upon TA treatment. (B) lncRNA expression levels were assessed via qRT-PCR in TA-treated and untreated stretched HSFBs. Data were normalized to GAPDH expression. (C) HSFBs were transfected with scrambled (si-control) or a LINC01569-targeted small interfering RNA (siRNA) (si-LINC) and then subjected to TA and stretch treatment. The medium of cultured cells was harvested and analyzed using ELISA. (D to H) HSFBs were transfected and treated as described in (C), and cells were subjected to qRT-PCR for MMPs (D), TUNEL staining (green) for apoptosis assay (E and F), CCK-8 assay (G) and Western blotting analysis at 48 hours after transfection (H). (I) Sections of hypertrophic scar tissues treated with or without (control) TA were prepared and subjected to ISH for LINC01569. For (C), (D), (F), and (G), data are presented as means ± SD. *P < 0.05 and **P < 0.01, by Student’s t test.
