Susceptibility and permissiveness of B cells to DENV infection in vitro. (A) CD14+ monocytes and CD19+ B cells were isolated from PBMCs from healthy donors by magnetic sorting and infected with reference or low-passaged DENV-1 and DENV-2 strains at MOI of 20. At 24 h post infection, the cells were stained intracellularly with anti-DENV NS3 antibody. Infectious viral progeny in supernatants from healthy donors (n = 6) infected with DENV-1 and -2 reference or low-passaged stains at MOI 20 for 24 h was determined by focus-forming assay. (B) Total DENV viral RNA copies in supernatants from healthy donors (n = 6) infected with DENV-1 and -2 reference stains at MOI 20 for 24 h was determined by RT-qPCR.
