A two-step procedure enables bioconjugation of IgG to mPAC. (A) The thiol-conjugation reaction with mPAC and linker peptide affords G₃-mPAC. (B) Sortase-mediated ligation with IgG antibody (IgG-LPSTGGK) and G₃-mPAC affords a mixture of IgG-mPAC and IgG-(mPAC)₂ conjugates. Coomassie-visualized SDS-PAGE gel analysis of mixtures containing: (C) Tmab and G₃-mPAC (linker peptide 1), before and after adding SrtA*; and Tmab-mPAC conjugate after (D) size-exclusion chromatography (SEC, HiLoad 16/600 Superdex 200) and (E) anion-exchange (AEX) chromatography (HiTrap Q HP).
