H₂O₂-dependent hyperoxidation of PRX-IIE. Pre-reduced and desalted PRX-IIE at 10 µM concentration was incubated with H₂O₂ as indicated in the presence of 500 µM DTT for 15 min. This allowed for continued re-reduction and H₂O₂-dependent oxidation and accumulation of the hyperoxidized form. The extent of sulfinic acid formation in percent of the total was estimated from the ratio of the peak intensities for the reduced (SH; 19.438 Da) and hyperoxidized (SO₂H; 19.470 Da) protein in the deconvoluted ESI-MS spectra. Data are means of n = 10 ± SD with recombinant protein from two independent purifications.
