Experimental plate designs for intra-plate variability. Peptide-12 (P): two 384-well plates were analyzed: PA1 (positive kD): A4.5 = acetate buffer, pH 4.5, and the concentration series were arranged horizontally; ascending concentrations are placed on the upper half of the plate and descending concentrations on the bottom half (1 = 2 mg/mL, 2 = 4 mg/mL, 3 = 6 mg/mL, 4 = 8 mg/mL, 5 = 10 mg/mL API, and 0 = placebo). The design of plate PP1 (negative kD) for phosphate buffer, pH 6.0, was equal to plate PA1.
