Clonogenic assay: (a) U2OS; (b) HOS. Cells were treated with ICOS-Fc, SD-Sr-ICOS-Fc, SD-Sr, SG-Sr-ICOS-Fc, and SG-Sr at 2 and 0.2 μg/mL concentration for 72 h. Then, the cell medium was changed, and the cells were cultured for additional 7 days in a free medium (c), (d) Colonies were then photographed. Then, the cells were treated with acetic acid to induce a completely dissolution of the crystal violet and absorbance was evaluated. Three different experiments were performed.
