Ad.5-TCTV inhibits cancer cell proliferation to a similar extent as Ad.5-CTV. (AD) The indicated cell types (5000 cells), (A) human breast cancer (MDA-MB-231 and SUM-159); (B) human prostate cancer (DU-145 and PC-3); (C) primary normal human breast epithelial (HMEC); and (D) normal immortal prostate epithelial (RWPE-1) cells were plated in 96-well plates and infected with either Ad.5-E1A (25 pfu), Ad.5-CTV (25 pfu), or Ad.5-TCTV (25 pfu) for 72 h. These cells were then treated with 20 μL of MTT in phosphate-buffered saline (PBS) and incubated further for 4 h. Media was then removed and formazan crystals were dissolved using dimethyl sulphoxide (DMSO) (100 μL). A microplate reader was used to measure the absorbance at 550 nm and the results are presented as percent proliferation with the comparison of cells treated with vehicle.
