Exacerbation of mechanical allodynia and upregulation of DRG Piezo2 expression by selective activation of Epac1 or NR2B in normal mice. Normal mice were randomly divided into three groups (n=8/group) and received intrathecal (i.t.) saline (5 µl) and NMDA (1 nmol/5 µl), or intraplantar saline (5 µl) and 8-pCPT (12.5 nmol/5 µl), respectively. (A) PWMT was measured at 0 (before injection), 5, 15, 30, 60 and 90 min after intrathecal or intraplantar injection. (B-D) The expression level of the right lumbar DRG L2-L5 segments of Epac1in 8-pCPT group, p-NR2B in NMDA group and Piezo2 mRNA in both groups were detected at the time of 0, 15 or 30 min after injection.
