Frozen PBMCs isolated from healthy adults were labeled for CD3, CD4, CD25, and CD127. Coexpression of CD25 and CD127 on gated CD3+CD4+ cells is presented in logical (A) and logarithmic (B) scale. (C) Isotype control staining for CD25 and CD127 expression is shown in red. For A–C, 127-hi cells are in the box in the top right quadrant and coexpress CD127 and CD25 (CD25+CD127hi), Tregs are in the box in the bottom right quadrant and express a low density of CD127 and a high density of CD25, and CD25– (CD25-neg) cells are in the top and bottom left quadrants. The data are representative of more than 10 healthy donor PBMC samples.
