(A) B6 mice were treated with mouse IFN-α, poly(I:C) dissolved in NaCl (PIC-NaCl), or poly(I:C) emulsified in lipid nanoparticles (PIC-LNPs) and sacrificed at 4, 8, 24, or 72 hours after treatment (n = 3 for each group and time point). The mice were also sacrificed at 24 hours after injection of NaCl or LNPs to serve as controls. The mRNA expression levels of Ifnb, Isg15, and Oas2 in the liver (left panels) and the spleen (right panels) were analyzed by RT-qPCR. (B) Hepatocytes and intrahepatic lymphocytes (IHLs) were isolated from the liver of B6 mice treated with PIC-NaCl and PIC-LNPs (n = 2 for each group) and analyzed for mRNA expression of Ifnb and Oas2. Data represent mean + SD (n 3).
