ELF3::HLA-C axis in EVTs. (A) The ELF3 gene is regulated by a superenhancer and an autoregulatory feedback loop. ELF3 reinforces its own gene expression by binding to its own promoter and recruiting the Mediator component MED23/SUR-2. Wrenchnolol disrupts the interaction of ELF3 and MED23/SUR-2, and (+)-JQ1 disrupts the superenhancer. (B) ELF3 regulates HLA-C expression. RFX5 competes with ELF3 for the binding to the ETS motif in the HLA-C promoter. The Mediator component MED1, BRD4, and other factors, together with ELF3, form the HLA-C enhanceosome and establish HLA-C expression at the maternal–fetal interface.
