HEK293 cells (1X107) were transfected with expression plasmids encoding the epitope tagged forms of the indicated proteins. 48 h post-transfection, cells were lysed and epitope tagged proteins were immunoprecipitated (IP) with anti-Flag beads, separated by SDS-PAGE and analyzed by western blotting (WB) using the indicated antibodies as described in Materials and Methods. (A) Co-immunoprecipitation of Myc-ZBTB2 and Flag-tagged HDACs 1–9 or (B) V5 tagged HDAC10. (C) Summary of ZBTB2 and cellular HDAC interactions. Interaction (+) was defined as having an IP efficiency greater than 4.5, weak interaction (+/-) as less than 4.5 and no interaction (-) as less than 1.5. (D) Co-immunoprecipitation assays of Flag-HDAC4 and Myc-WT ZBTB2 or Myc-ΔPOZ ZBTB2. IP efficiency is the ratio of % starting material immunoprecipitated by HDAC verses the % starting material precipitated with no HDAC (e.g. ratio of lane 3:lane 2 in A and B).
