Myo9b GAP and motor activities regulate macrophage motility. Mutant Myo9b constructs deficient for either Myo9b GAP or Myo9b motor activity are unable to rescue migration velocity and directionality of Myo9b-deficient HL-60 macrophages. Indicated HL-60 cells differentiated to macrophages were subjected to random migration on fibronectin-coated slides, and individual cells were tracked to calculate velocity (AB) and persistence (directionality) (CD) of cell migration. Data are from at least three independent experiments with 10 cells tracked each. A p-value of p ≥ 0.05 was regarded as not significant (n.s.), p ≤ 0.05 as a trend (∗), p ≤ 0.01 as significant (∗∗), and p ≤ 0.001 as highly significant (∗∗∗).
