NHE inhibition impairs Isc recovery in response to hypertonic NaCl and NHE2-KO lowers basal Isc. Wild-type (Control, C57BL6/J), or NHE2-WT and NHE2-KO (FVB/N) tissues were mounted into an Ussing chamber and WT challenged with 0.5 M NaCl at t = 10 min. Results compared Control tissue in the absence (black) versus presence (blue) of 20 μM HOE 694 added at t = 40 min (arrow) to both luminal and serosal bath, measuring a representative time course of Isc, b percent (%) Isc recovery 2 h after 0.5 M NaCl, or c change in (Δ) transepithelial R 2 h after 0.5 M NaCl.
