6BN has high potency to prevent opioid withdrawal in adult guinea pigs. Animals were treated for 3 days with one injection per day of 10 mg/kg MTD with varying doses of 6BN (0, 0.01, 0.03, 0.3, and 1 mg/kg). On day 4 animals were injected with naloxone and tested for 30 min in an open-field arena. (A) 6BN delivered at low doses with MTD prevents naloxone-induced withdrawal in adult animals. p < 0.05 for overall effect of 6BN, by linear trend test. (B) Same as A, but three animals with >15% of total arena-time in hypotonic posture were removed from dataset. p < 0.01 for overall effect of 6BN, by linear trend test. (A), (B) *p < 0.05, **p < 0.01, ***p < 0.001, compared to “0 6BN” control, by Dunnett post-hoc test; ψp < 0.05 compared to saline control (gray lines, mean ± SEM), by t-test.
