Figure 7. The cardioprotective effect of preconditioning with exoCDNF is blocked by an anti‐CDNF antibody or by CDNF‐ΔKTEL.

Figure 7

CDNF (1 µmol/L) or CDNF+antibody were perfused before ischemia (5 minutes) (A, B, and E). CDNF‐ΔKTEL was unable to confer cardioprotection to isolated perfused hearts subjected to I/R. Isolated perfused hearts were perfused with CDNF‐ΔKTEL (1 µmol/L/5 minutes) before I/R (C, D, and F). Time courses of (A and C) left ventricular developed pressure (LVDP) and (B and D) left ventricular end‐diastolic pressure (LVEDP) during I/R protocol. Representative cross‐section images of TCC‐stained ventricles hearts subjected to I/R in the absence or in the presence of CDNF, CDNF+anti‐CDNF antibody (E), or CDNF‐ΔKTEL (F). Number in each column is n of hearts. The data were expressed as means±SEM. @ P<0.001 vs control; *P<0.001 vs preCDNF. With 1‐way ANOVA followed by Bonferroni post hoc tests for infarct area analysis and 2‐way ANOVA followed by Bonferroni post hoc tests for LVDP and LVEDP analysis. CDNF indicates cerebral dopamine neurotrophic factor; isch, ischemia; rep, reperfusion; and TCC, triphenyltetrazolium chloride.
