The overall workflow of single-cell sequencing (SCS). Single cells are isolated from solid tissues or liquid biopsies via different approaches. Libraries generated from purified nucleic acids are sequenced on the 2nd or 3rd generation sequencing platforms. Raw reads are processed and visualized as shown in different applications in the Analysis section, including marker gene selection by differential expression, pseudotime tracking by trajectory analysis and cell type cluster to elucidate the population. Parts of the subplots are adapted from the figures in the reference literature [11,12,13]. CTC: circulating tumor cell; DTC: disseminated tumor cell.
