Triptolide treatment induces autophagy in SH-SY5Y neuroblastoma cells. (A) Investigation of SH-SY5Y LC3 protein levels by Western blotting showed increased expression of LC3II in triptolide-treated cells at 6 and 24 hours, which is consistent with autophagy. Actin served as a loading control; results representative of n =3. (B and C) Treatment of SH-SY5Y cells with triptolide at 100 nmol/L for 20 hours shows a significant increase in the LC3 punctate staining pattern when compared with untreated controls. This finding was not observed in the apoptotic IMR-32 neuroblastoma cell line. Data are expressed as the mean ± SEM of 3 independent experiments.
