Nimbolide altered the expression of proteins associated with actin dynamics

(A–C) Immunofluorescence images of MDA-MB-231 cells treated with nimbolide shows the localization of (A) F-actin and pVASP, (B) F-actin and Diaph1, and (C) F-actin and phosphorylated p21 activated kinase (pPAK). (D) Nimbolide-induced actin depolymerization is overcome by Rac1/Cdc42 activation. MDA-MB-231 cells were treated with 1 μg/mL of Rac1/Cdc42 activator (bacterial cytotoxic necrotizing factor [CNF] toxin-based G-switch reagent) for 1 h and followed by nimbolide for indicated times. The cells were analyzed for F-actin and pPAK expression using immunofluorescence analysis (n = 3).
