Schematic of the vectors used in this study

The integrated forms of the HIV-based lentiviral vectors are shown. Two versions of vectors expressing wild-type (WT) cystic fibrosis (CF) transmembrane conductance regulator (CFTR), codon-optimized CFTR version 3 (coCFTR3), or GFP were compared. (A and B) Only the promoters used to drive transgene expression differ between the two versions, either (A) human phosphoglycerate kinase (PGK), or (B) human elongation factor 1-α (EF1α). A human ankyrin 1 (Ank) insulator element in the reverse orientation is present within the self-inactivating (SIN) 3′ LTR. Vectors shown are not to scale.
