Codon optimization of CFTR does not change CFTR single channel properties
Patch-clamp studies were performed using inside-out membrane patches from HEK293 transiently expressing WT CFTR or coCFTR3. (A) Representative tracings are shown. PKA catalytic subunit (22 nM) and ATP (1 mM) were present throughout. Holding voltage was −70 mV. For illustration purposes, traces were digitally low-pass filtered at 50 Hz. c, closed state; o, open state. (B−D) Mean channel open probability, burst duration, and interburst intervals were not different between WT CFTR and coCFTR3. Mean ± SE is shown.
