Summary algorithms for constructing two-parameter (T,D0) equi-dosing regimen regions

Analytical approach (one- and two-compartment models) Numerical approach for TCM (construct flo(T) and fhi(T) numerically)

Find expression for max and min values of ac, namely ac,max and ac,min, in terms of dosing regimen parameters D0 and T, as in (5.2) and (5.5)

Set therapeutic range constraints Dme<ac,min and ac,max<DMS

Rearrange constraints into form flo(T)<D0<fhi(T), as in (5.3) and (5.6)

Plot curves D0=flo(T) and D0=fhi(T). Region bounded by these two curves is the EDRR

Discretise the (T,D0) parameter space, i.e. lay down a grid of points (Ti,D0,j)

For each Ti:

   For each D0,j:

Use expression for ac(t) (3.28) to compute ac,max and ac,min numerically

   End

flo(Ti)=D0,j such that |ac,min-Dme| is minimised

fhi(Ti)=D0,j such that |ac,max-DMS| is minimised

End

Plot curves D0=flo(T) and D0=fhi(T). Region bounded by these two curves is the EDRR
